Relation between 2-deoxyglucose phosphorylation and adenine nucleotide content in Lettré ascites cells.

چکیده
The relation between the phosphorylation of deoxygluoose (2-DG) and levels of adenine nucleotides was investigated in endogenously respiring Ehrlich Lettréascites cells pretreated with various inhibitors. Pretreatment with the respiratory inhib itor, rotenone, caused a 70<; decrease in 2-deoxyglucose-6phosphate formation. The sources of energy for 2-DG phos phorylation in the presence of rotenone were found to be endoge nous reserves of adenine nucleotides, as well as a limited residual respiration. Phosphorylation of 2-DG in the presence of 10~4M dinitrophenol was 70' ; of normal, and was abolished by addition of rotenone. Dinitrophenol was shown to induce a stoichiometric transamination of alanine to pyruvate, and cause a marked increase of >4CO-> and pyruvato-14C from alanine-14C or glutamic acid-I4C. The evidence suggests that substrate level phosphoryla tion from a-o\oglut arate and subsequent formation of adenosine triphosphate by a decarboxylation reaction is at least partially responsible for the phosphorylation of 2-DG in uncoupled cells.

certain conditions this delicate balance may be disrupted. For example, during massive ATP dephosphorylation and impaired glycolysis [such as that which follows the addition of 2-deoxyglucose (7), glucose plus iodoacetate (16), or glucose plus oxamate (9)1 , there is an elevation of and then a decrease in AMP and ADP. This is followed by a decrease in AT? and AXP.4 The disruption of the energy ...

The phosphorylation of 2-deoxyglucose by respiring Krebs-2 ascites tumor cells and the effect of 2-deoxyglucose metabolism on cell orthophosphate, acid-labile phos phate, and adenine nucleotide-phosphate in these cells was investigated. In the presence of 2.0 mM orthophosphate 2-deoxyglucose (2DG) was taken up initially very rapidly, 5.7 /umoles/min/ml of cells, but uptake fell off to a steady ...
متن کاملOxidative Phosphorylation and Ultrastructural Transformation in Mitochondria in the Intact Ascites Tumor Cell
We have examined the ultrastructure of mitochondria as it relates to energy metabolism in the intact cell. Oxidative phosphorylation was induced in ultrastructurally intact Ehrlich ascites tumor cells by rapidly generating intracellular adenosine diphosphate from endogenous adenosine triphosphate by the addition of 2-deoxyglucose. The occurrence of oxidative phosphorylation was ascertained indi...

Experiments were conducted to determine whether en ergy charge, (ATP + 1/2 ADP)/(ATP + ADP + AMP) (in which AMP, ADP, and ATP are adenosine mono-, di-, and triphosphate, respectively) or intracellular inorganic phosphate was the primary regulator of the rate of aden osine 5 -monophosphate (AMP) catabolism in intact mouse ascites tumor cells. Hyperdiploid (ELD) and tetraploid (ELT/B1) cell suspe...
متن کاملAdenine Nucleotide Degradation during Energy Depletion in Human Lymphoblasts
Adenine nucleotide breakdown to nucleosides and purine bases was measured in cultures of human lymphoblastoid cells following: 1) the inhibition of oxidative phosphorylation in the absence of glucose or 2) the addition of 2-deoxyglucose. A mutant cell line, deficient in adenosine kinase, in the presence of an adenosine deaminase inhibitor was used to measure utilization of the two pathways of A...
